Formulation and In-Vitro Evaluation of Colon Targeted Indomethacin Tablet Using Tamarind Seed Polysaccharides by 32factorial Design

Abstract : In present study, an attempt was made to formulate colon targeted indomethacin tablet using tamarind seed polysaccharides and ethyl cellulose as release modifier and coated using Eudragit L100 and PEG 400 as plasticizer. 32 factorial design was applied to study the effect of number of polymers (tamarind seed polysaccharides and ethyl cellulose) and extend of Eudragit L100 coating % as independent variables on the dependent variables are Q4 and Q12. The tablets were analyzed for their hardness, friability, weight variation and an in-vitro drug release and drug release kinetics were performed in acid buffer 1.2 for 2hrs, in phosphate buffer 6.8 for 3hr and phosphate buffer 7.4 for 7hrs.The results indicated that Eudragit L100 and PEG 400 enteric-coated matrix tablets were successful in achieving gastric resistance and timed-release of the drug, assuring an adequate lag time for the intended colonic targeting, followed by a controlled-release phase was achieved by tamarind seed polysaccharides and ethyl cellulose. The drug release varies with concentration of polymer in tablets.
